• Microbiological and biochemical knowledge with a special focus on gram negative bacteria, recombinant proteins, vesicles
• Basic expression and purification techniques
• Ability to make molarity calculations for the preparation of solutions and buffers and for diluting, quantifying and dispensing proteic material

What will you do?

Context

Fermentation, protein purification and analytics team (FPPA lab) operating within the Antigen Design function in Research department is mainly involved in the production, purification, analytical and functional characterization of novel proteic vaccine candidates with the perspective of process transfer at manufacturing scale.

The intern will initially be supported by experienced scientists of the laboratory to learn the fermentation and chromatographic techniques necessary for the production and the purification of the antigen of interest.

Project intern description

The project has the objective to exploit the antigen expression profiles in different bacterial compartments with a special focus on membrane extracts from a subset of bacterial strains cultured with different media/conditions/nutrients. The candidate will evaluate the effect of different media, nutrients and co-factors on the expression and localization of specific antigens in E. coli and Klebsiella pneumoniae.

Subsequently he/she attempt the enrichment or the purification of target antigens for analytical characterization and immunogenicity studies.

Competencies the intern will develop thanks to the internship experience:

Learn and understand the techniques of fermentation, purification, and basic analytical characterization of proteic antigens. In particular, the student will learn how to grow gram negative bacteria in flask and in bioreactor, affinity and hydrophobic chromatography, SE-HPLC, SDS-page, western blot, quantification methods. All these techniques will allow him/her to produce and characterize antigens that can be used as vaccine candidate.
